Melanotan II (MT-II) is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone (alpha-MSH), developed at the University of Arizona as part of melanocortin receptor research. It acts as a non-selective agonist at MC1R, MC3R, MC4R, and MC5R, with particularly high potency at MC1R, the receptor governing melanogenesis in epidermal melanocytes via the cAMP-PKA-MITF pathway. It also activates central MC4R, the basis of its documented effects on arousal and appetite in animal studies, and was the precursor compound from which the more selective MC4R agonist Bremelanotide (PT-141) was derived.
